Recessive gene disorder: Shared descent from a common ancestral population plays a bigger role than consanguinity for many recessive gene disorders

A paper published online today (September 24) in the journal Nature, shows that all diverse groups seen spread out in India today come from two major ancient populations that are genetically divergent.

The two ancient populations are the Ancestral North Indians (ANI) and the Ancestral South Indians (ASI). The study was based on genetic analysis.

While the Ancestral North Indians (ANI) group is genetically close to Middle Easterners, Central Asians, and Europeans, the Ancestral South Indians (ASI) are not related to any group outside India, notes the paper.

வடக்கிந்திய ஆதி மக்கள் மரபணு சோதனையில் அவர்கள் மத்திய கிழக்கினரையும், மத்திய ஆசிய மக்களையும், ஐரோப்பியர்களையும் ஒத்திருப்பதாகவும், தென்னிந்திய ஆதி மக்கள் எந்த ஒரு வெளி நாட்டு மக்களோடும் சார்ந்திருக்கவில்லை என்பதையும் இந்த ஆராய்ச்சி தெளிவு படுத்துகிறது.

The study was undertaken by the Hyderabad-based Centre for Cellular and Molecular Biology (CCMB) and three other institutions in the U.S.

Samples from 132 people representing 25 groups from 15 States and speaking six language families (including two language families from the Andaman Islands) were studied.

The study also looked for genetic variations based on caste — upper and lower caste — from two the States of Uttar Pradesh and Andhra Pradesh.


The study found the groups (seen today) that emerged from the two ancient populations have distinct genetic affinity. “The populations (groups) that emerged from ANI show 40-80 per cent genetic affinity to European population.

Senior Scientist at CCMB, and one of the authors of the paper. “The Hyshi and Ao Nage population from north-east India show genetic affinity to the Chinese.”

The indigenous population seen in the Andamans have more affinity to the ASI. “Otherwise, they have no relationship with any other population anywhere in the world,” said Dr. Thangaraj.

Unlike the European and Chinese population, the Indian population are more scattered in a genetic sense.

Medical implications

While consanguinity is often implicated for many recessive gene disorders in a population, this study found that the “shared descent from a common ancestral population plays a bigger role. This is called as the ‘founder effect.’

Founder effect is nothing but the fact that many groups seen today have descended from a small group of founding individuals, and these founding individuals in turn have been isolated from other groups, genetically speaking.

It is based on this fact that the authors state that the founder effect plays a bigger role. “We propose that the founder effect is responsible for an even higher burden of recessive diseases in India than consanguinity,” the paper states.

Dr. Thangaraj explained the significance of this. A disease can occur due to the presence of a recessive gene due to mutation. In a small population with high endogamy [where people marry within the population], the mutation persists and spreads to more number of people.

After a point of time, a large number of people have the recessive gene and this increases the chance of a child receiving a recessive gene from both the parents and thus becoming diseased.


Existence of caste

Contrary to popular perception by historians that the caste system seen today is an invention of colonialism, the study found scientific evidence to show that “many current distinctions among groups are ancient.”


“The caste system is not recent,” said Dr. Thangaraj. “The social stratification existed right from early human divergence, some 50,000-60,000 years ago when initial settlement happened in India.”

The paper adds a word of caution: “Models in population genetics should be treated with caution. Although they provide an important framework for testing historical hypotheses, they are oversimplifications.”

leaving aside other comments, for your anxiety let me tell two things. sample is not just 132 individuals.the actual data is as follows:

secondly, hope you know the academic level of Nature. so people doing work for such a journal may not have gone for sample errors.

Once upon a time 'The Hindu' was also a very highly respected newspaper (still highly respected by some people) - does that mean we need to agree everything published in 'The Hindu'. If some controversies arise, Nature will also put a disclaimber saying 'these are the opinions of the authors of the article and not of Nature'. Correct thaanE aiyaa? :-)

The sample data statement is a bit confusing. Whether it is saying that the sample consists of just 132 individuals or it consists of 132*25 individuals or it consists of 132*25*13 individuals? If it is just 132 individuals only but they are from 25 diverse groups from 13 different states, my comment is still valid. 132 individuals is a very small (even can be said negligible) number compared to the population size.

I have read other research results where just one virumaandi's genome was compared with African genome, and declared affinity between Africans and South Indians. This is repeatedly quoted by many munaivars highly respected by me - but still I question the nature of this research.

May be I am reacting as a layman here. All these Doctorates consider this size of sample good enough to prove these kind of research results. Only the Academia know. (I did not say my natural comment 'Only the God knows' here):-)

A new research has revealed that nearly all Indians carry genomic contributions from two distinct ancestral populations. Following this ancient mixture, many groups experienced periods of genetic isolation from each other for thousands of years.

The study, which has medical implications for people of Indian descent, was done by scientists at the Hyderabad-based Centre for Cellular and Molecular Biology (CCMB) together with US researchers at Harvard Medical School, the Harvard School of Public Health and the Broad Institute of Harvard and MIT.

The results of the study were disclosed to the media here on Thursday by CCMB director Veena K Patnaik, its former director Lalji Singh and scientist K Thangaraj.

The study revealed that many groups in modern India descended from a small number of founding individuals, and have since been genetically isolated from other groups. In scientific parlance, this is called a ‘founder event’.

According to Thangaraj, this also has important medical implications. “The widespread history of founder events helps explain why incidence of genetic diseases among Indians is different from the rest of the world,” he said.

To find out the genetic variability across the Indian subcontinent, the research team analysed more than 500,000 genetic markers across the genomes of 132 individuals from 25 diverse groups spread across 13 states.

The genomic analyses revealed that different Indian groups have inherited 40 to 80 percent of their ancestry from a population that is now called ancestral north Indians, who are related to western Eurasians, and the rest from the ancestral south Indians, who are not related to any group outside India.

1.மனித இனம் ஆப்பரிக்காவில் தோன்றிய பின்பு முதலில் இந்தியாவிற்கு வந்தோர் ஆஸி- இந்திய குடியினர், இது 60,000 வருடங்களுக்கு முன்பு.

2. கிபி 3000-ல் வந்தவர்கள் திராவிடர்கள். கவனிக்க ~57000 வருடத்திற்கு பிறகுதான் திராவிடர் வந்துள்ளனர். ஆஸி-இந்திய குடியினர் திராவிடர் படையெடுப்பால் சிறுகுழுக்களாக சிதறினர்.

3. கிபி 1500-ல் இந்தோ-ஐரோப்பிய ஆரியர்கள் வந்து திராவிடர்களை தெற்கே விரட்டினர்.

ஆக நாம் அனைவரும் வந்தேறிகள்தான்...ஆல் கைபர் போலன் பார்ட்டிகள்தான்!

மேலும் nature-ல் வந்தால் அது சரிதான் எனும் தருமி சாரின் கருத்தில் எனக்கு உடன்பாடு இல்லை.


ஆனால் ஏற்கனவே Indian Genome Variation Consortium (IGVC) 1871 பேரை வைத்து செய்த ஆராய்ச்சியின் அடுத்த கட்டம்தான் இது. (IGVC)75 ஜீன்களிலுள்ள 420 இடங்களை மட்டும் ஆராந்தனர்.தற்போது இவர்கள் 132 பேரின் முழு DNA வையும் (5,60,123 இடங்களை) பிரித்து மேய்ந்து விட்டார்கள். இதனால் சாம்பிள் எண்ணிக்கை அதிகரிப்பது கடினம்.

தென்னிந்தியர்கள் தனிச்சு நிற்பதால் உடனே இந்தியாவில் மனிதன் தோன்றினான் என எடக்காக முடிவு செய்யகூடாது.

ஆப்ரிக்காவில் மனிதன் தோன்றி பல ஆண்டுகளுக்கு பிறகு மனித குழுக்கள் மற்ற கண்டங்களை நோக்கி கிளம்பின. அப்போது இந்தியா ஒரு hub ஆக செயல்பட்டதாக ஆராய்ச்சியாளர் கருதுகிறார்கள்.

ஐரோப்பிய-வடஇந்திய மரபணு ஒற்றுமை பல வழிகளில் சாத்தியம்.ஐரோப்பிய குரூப் ஆப்பரிக்காவிலிருந்து ஐரோப்பிய போய்விட்டு இந்தியா வந்திருக்கலாம். அல்லது இரண்டாக பிரிந்து ஒன்னு ஐரோப்பியாவிற்க்கும் இன்னொன்னு இங்கும் வந்திருக்கலாம். அல்லது பெரும்பாலோனோர் நம்புவது போல் இந்தியா வந்துவிட்டு ஐரோப்பா போயிருக்கலாம்.

ஆனால் இன்னோர் குழு (ASI) கிளம்பி மொத்தமாக இந்தியா வந்துவிட்டால் அவர்களுடன் வேறு யாருக்கும் தொடர்பு இருக்காதில்லையா?

மேலும் ANI மற்றும் ASI இருவருமே ஆப்ரிக்க இனத்தவருடன் நெருஙகிய தொடர்பு இல்லாமலிருக்கிறார்கள். இதற்காக மனிதன் ஆப்ரிக்காவில் தோன்றவில்லை என புரிந்துகொள்ளக்கூடாது.

நான் எனது நண்பனுடன் இரண்டு வருடத்துக்கு முன்பு அனுமானத்தில் விவாதம் செய்தேன்.இன்னும் சில வருடங்களில் உண்மை ஆகிவிடும் போல இருக்கு.ஆனால் இந்த ஆராய்ச்சிக்கு இந்தியாவின் தலை ஈடும் அதிகமாகவே இருக்கும் என்று தோன்றுகிறது.
